Mouse liver-specific infection with avia retroviruses - a mouse model to dissect the multistep processes of hepaticcellular carcinomas

Objective



Insertional mutagenesis has been used in combination with transgenic mice carrying activated oncogenes or lacking functional tumor suppressor genes, in order to focus the studies on multisteps processes tumorigenesis in particular cell lineage. This approach succesfully demonstrated oncogenes collaboration in lymphonagenesis.
The goal of this proposal is to develop a transgenic mouse model carrying tissue-specific infection susceptibility to subgroup-A avian leukosis-sarcoma retroviruses (ALSV), to study multistep processes in tumorigenesis of hepatocellular carcinomas. First. A model will be developed to target lymphoid compartment in order to test the in vivo infectibility of mouse tissues with ALSV and their subsequent tumorigenic-capacities. Second. This model will be used to direct mouse liver-induced tumor. These mouse strains should be mated with previously developed transgenic mice strains with liver-specific oncogene expression or heterozygotes, homozygotes KO mice strains or chimera mice for tumor suppressor gene which are now available, to identify events implicated in hepatocellular carcinogenesis.
