Analysis of the action of enod40 on plant differentiation and growth control

Objetivo

Msenod40 is expressed in differentiating cells from the nodule primordi and in spontaneous nodules of Medicago plants It is proposed that this genes might act as "riboregulators", a recently described class of untranslated RNAs involved in differentiation, and we are now analysing their role on nodule morphogenesis and other differentiation processes.
We propose to construct transgenic plants that overexpress the enod40 gene in sense and antisense orientation using constitutive, inducible and/or tissue specific promoters, to analyze its role "in planta". In parallel, using rapid bioassays, structure-function studies will be performed on the Mtenod40 RNA to delimit biologically active regions involved in cell proliferation. This will allow to conclusively answer if any of the small ORFs can indeed be transated or if a novel function the RNA in the cell may be stablished in relation with plant differentiation.
