Objective
The biosynthesis of glycoproteins bearing N-linked oligosaccharides is accompanied by the release of free polymannose oligosaccharides into the lumen of the endoplasmic reticulum (ER). Recently I have shown that these oligosaccharides are transported out of this compartment into the cytosol. The aim of the project described herein is to characterize this ER-to-cytosol transport process for free oligosaccharides at the molecular level. Using permeabilised cells, microsomes, and inverted red blood cell/microsome chimeras I will obtain the first data concerning the molecular nature of this transport process including information relating to its specificity, saturability and kinetic properties. These important data will provide the basis for the purification strategy that will be undertaken in order to more fully characterize the transporter molecule. Understanding the mechanism of the ER-to-cytosol transport of free polymannose oligosaccharides will ultimately help identify the physiological purpose of this recently described process.
