A functional approach by t-dna tagging to isolate plant genes involved in salt tolerance

Objective



Saline stress in agricultural crops is a major challenge to plant biotechnology. Although a genetic background to stress tolerance exists in halophytes, this is not available to breeders of crop plants. An application of biotechnological advances requires the knowledge of target genes responsible of salt tolerance.
Previous work with the baker's yeast S.cerevisiae has revealed the use of a 'functional approach', that is gene overexpression and selection of those activated genes which confer the expected phenotype, as a very efficient procedure to identify genes capable of increasing salt tolerance. Obviously, a straightforward advance would require the use of a similar 'functional approach' of overexpression and selection directly in plants. The recently designed T-DNA tagging procedure combined with protoplast cocultivation allows the isolation of screenable dominant mutations by overexpression of any particular gene which can be later rescued.
In the present project we propose utilize this innovative strategy with tobacco protoplasts to identify plant genes involved in salt tolerance.
