Objective
The plant reproduction process plays a key role during the life cycle of the plant. Characterization at the molecular level of flower development and seed formation is essential for a beKer understanding of these basic processes as well as for the development of methods for the manipulation of these processes.
Transcription factors belonging to the MADS box family will be isolated from Zea mays ovule and zygote specific libraries. A Mutator transposable element system will be used for the functional analysis of these isolated genes. A new method based on screening of insertion events of mobile genetic elements into genes of known sequence by PCR, has been proposed for the production of mutants. The phenotype of these mutants will give a clue on the function of the disrupted genes. This site selected transposon mutagenesis system will be used to study the function of genes involved in ovule and seed development in Zea mays.
The mutants obtained will be characterized morphologically by Light Microscopy and Scanning electron Microscopy and molecularly by in situ hybridisation. This approach will lead to the identification of new genes important for ovule and zygote development in Zea mays.
