Objective
The applicant proposes a research project aiming to integrate genetic linkage and chromosomal maps of cultivated beet Beta vulgaris, a crop species of major agronomic importance in Europe. Genome analysis impacts strategies for gene isolation and expression, and crop improvement by genetic engineering. The proposed projects is targetted to highlight key features of gene-enriched regions of the genome of cultivated beet. The correlation of genetic and physical sugar beet maps will be achieved by interrelated methods using 1) molecular fluorescence in situ hybridization and high resolution image generation and processing; 2) micromanipulating techniques to dissect and isolate single Beta chromosomes which will be either tested in PCR-based assays or used for the construction of chromosome-specific DNA-libraries; 3) Laser-based flow sorting of native and PRINS differentiated Beta chromosomes as a starting point for establishing a set of chromosome-specific libraries covering the sugar beet genome. Emphasis is also given to a comparative molecular cytogenetic study on heterochromatin synteny between cultivated and wild beet species.
