A gene trap screen for the identification of genes involved in motoneuron development

Objective



My aim is to dissect the molecular events leading to the differentiation of a specific sub-class of neurons i.e. motoneurons, by using the recently-developed gene trapping technology to identify genes which are specifically transcribed in cells which are differentiating toward a motoneuron phenotype.
1. A replicate bank of embryonic stem cell clones will be generated containing random integrations of the B- galactosidase reporter gene into the mouse genome.
2. Individual ES cell clones will be induced to differentiate in vitro towards a motoneuron phenotype using conditions recently identified in the host laboratory. Clones of interest will be identified by a B-gal assay and further characterised using molecular genetic techniques.
3. These clones will be used to generate transgenic mice in which the in vivo expression pattern of the B-gal (and thus of the trapped gene) as well as the effects on motoneuron development of null mutations in the trapped genes can be analysed.
