Objective
The NMDA subtype of glutamate receptors plays a critical role in neuronal development and function, and has been identified as an essential component in LTP, an intensely studied cellular correlate of learning and memory. The gene for the major subunit NR1 was deleted in mice through homologous recombination techniques, but mutant mice die on the day of birth. The current project aims at rescuing the phenotype of cells lacking NMDA receptor function by a chimaera approach: ES cells containing the double NR1 knockout are used to generate chimaeric mice. Double knockout cells in the brain can be identified by virtue of a marker gene (lacZ. These cells can be analysed by morphological criteria, antigen expression patterns, in situ hybridisation techniques, and whole cell and single channel recordings to better understand the consequences of lacking NMDA function in important parts of the brain, like hippocampus, striatum, cortex and cerebellum.
