Effects of external signals on gene expression in a model suspension culture system

Objective



Effects of external signals on gene expression during wound and stress responses in plants will be studied. Tomato suspension cultures will provide a useful system for studying the expression of annexins and 14-3-3 proteins, both on the gene and protein level. The annexins and the 14-3-3 proteins are involved in the signal transduction in eucaryotic cells by acting as targets for calcium signalling. Professor Bowles was the first to identify annexins and has shown that the tomato suspension culture system expresses all members of the two gene families. To further study how external signals influence the expression of annexins and 14-3-3 proteins, specific probes based on the cloned gene families of annexins and 14-3-3 proteins, will be used for Northern blotting. Antisera will also be prepared to identify the presence of the different members of the annexins and the 14-3-3 families at the protein level. The experience acquired will later be applied to suspension cultures of Norway spruce, a key economic species in Northern Europe.
