Molecular genetic analysis of pathogenicity of botrytis cinerea. Characterization of two in planta induced genes

Objective



The infection process of Borrytis cinerea on tomato is being investigated. As a part of this research fungal genes differentially expressed during pathogenesis are being isolated by "differential display of mRNA". The proposed research will focus on the analysis of two of these genes. A high level of expression in planta for the first one and differential expression in planta for the second one has already been demonstrated. The role of these two genes during pathogenesis will be investigated. To this end the genomic copies will be isolated and the gene structures determined. Their expression patterns in planta will be studied by Northern blot analysis and promoter-reporter gene (GUS) fusions. Whether these genes play an important role in pathogenesis will be determined by constructing mutant strains and assessing their pathogenicity in comparison with the wild type.
