Objective
We aim to develop the study of the reaction dynamics in enzyme active sites after photoinitiation of an active co-factor. Femtosecond studies are ideally compatible with pulsed synchrotron x-ray measurements now possible on the time-scales of nanoseconds to tens of picoseconds. This presents us with the opportunity to study dynamics which can be used to interpret structural information. The dynamics, we believe, will in general consist of fast events driving slower large-scale processes in a coupled manner as seen in the haemoglobin and rhodopsin systems. We shall compliment these experiments with related studies on model systems supported by theoretical calculations to aid interpretation of the data. The experimental techniques we shall use are those of femtosecond spectroscopy which will be easily extended to ultraviolet (around 200 nm) and also to the infra-red roughly 500 to 3000 cm-l for probing transients. Femtochemistry is a new and exciting area of science, the possibilities are endless, the opportunity to work and develop expertise in this area would, I feel, be particularly rewarding.
