Reconstitution of the mitochondrial outer membrane translocation machinery

Objective



Mitochondria synthesize very few of their constituent proteins. The majority of the proteins is nuclear-encoded and has to be imported into the organelle after synthesis on cytoplasmic ribosomes. Translocation of preproteins across each of the two mitochondlial membranes involves numerous components, including a membrane embedded translocation machinery. The outer membrane translocation machinery is comprised of the so-called MOM-proteins.
This proposal is dedicated to the in vitro reconstitution of the protein translocation complex of the mitochondrial outer membrane. The reconstituted system will facilitate the functional characterization of the individual MOM-proteins and the specification of the putative role of the anionic membrane lipids in the mitochondrial outer membrane in protein translocation. Furthermore, it will allow the identification of the recognition sites of preproteins during the translocation process and it will establish the minimal requirements for the transport reaction. My activities bring together two laboratories with quite different expertises, i.e. mitochondrial protein import and biochemistry of membranes and phospholipids. The combined technical support from these two laboratories should enable me to gain new insights into the functional roles of proteins and lipids during the translocation process.
