Hla-e transgenic mice - an experimental model for the in vivo study of the functions of hla-e molecules

Objective



The objective of our project is to set up an experimental model allowing the in vivo study of the functions of HLA-E molecules. With this aim, we have produced eight HLA-E transgenic mouse lines. The research programme will study: (i) the expression of HLA-E molecules, (ii) their role as transplantation antigens (skin graft rejection, bone marrow transplantation and graft versus host reaction), (iii) their antigen presentation capacity, (iv) their immunogenicity for B and T cells, (v) their influence on tumor growth, i.e. their possible role in immunosurveillance. In addition, the model opens up the possibility to produce HLA-E specific monoclonal antibodies, an essential and still missing tool for the detection of these molecules.In summary, the results of these experiments should contribute to the elucidation of the physiological significance and the role in transplantation of the abundant family of human nonclassical class I molecules.
