The characterisation of a putative human cell division cycle gene and study of its possible role in intestinal differentiation

Objective



During my present research on the intestinal enzyme lactase I have found a putative human cell division cycle gene. I wish to follow up this initial finding, and isolate and characterise a complete cDNA corresponding to this gene. This will enable the analysis of the relationship of this transcript to putative homologues identified in yeast and other organisms. I propose to investigate the pattern of expression using RT PCR, Northern blotting and in situ hybridisation techniques.
I also aim to make specific antibodies for analysis of the resulting polypeptide. I will study cell lines which are being used as models for intestinal differentiation in addition to cell lines in which the cell cycle call be regulated by drug treatments.
I also aim to clone and characterise the promoter region of this gene to enable future studies on the regulation of expression.
