Characterization of the chlamydia containing vacuole and its interaction with other intracellular compartments

Objective



Chlamydiae are obligate intracellular bacteria occupying a vesicle called an inclusion throughout their life cycle. They evade lysosome destruction by inhibiting fusion of the inclusion with lysosomes. There is very little information on the origin of the vacuolar membrane surrounding the chlamydiae or the properties of the compartment in which they proliferate. Additionally, little is known about the role of the cytoskeleton in chlamydiahost cell interaction. My proposed project concerns the intracellular survival of chlamydiae. The subcellular compartment(s) in which chlamydiae survive will be characterised by 1) following the distribution of specific intracellular organelle markers of the host cell in the chlamydiae compartment(s) and 2) measuring the pH of these vacuoles. In addition, the mechanism whereby chlamydiae avoid lysosomal destruction and the role of cytoskeletal elements in the formation of the chlamydia inclusion and the chlamydia infectious cycle will be investigated.
