Acylase aa-i as catalyst for irreversible acylation reactions - mechanistic studies and synthetic applications

Objective



The goal of this project is to develop and exploit enzyme mediated reactions for the synthesis of peptides and peptidomimetics. This class of molecules are becoming attractive synthetic targets because of their pharmacological potential. A promising biocatalyst is acylase I from Asperigillus species (AA-I), since it exhibits excellent selectivities in recently studied acylation reactions of amines by vinyl acetate. This project includes kinetic and mechanistic studies on the biocatalytic \ potential of AA-I. On knowing the biochemical capabilities of AA-I a methodology will be developed for the creation of peptide strands. The scope of AA-I will be extended to the synthesis of a set of building blocks containing non-proteinogenic alpha-amino acids to induce restricted conformations in the peptide strand.
