Research project on high field nmr and molecular. Dynamics study of interaction between dna and modified hoechst 33258

Objective



Double stranded DNA, the carrier of genetic information, is an ideal target for the treatment of cancer and viral desease. Inhibited gene expression by creating irreversible damages on DNA is now a widely use strategy.
Various nuclease chemical systems are described in literature. For example, mesotetrakis(4-N-methyl-pyridiniumyl)porphyrinato-Mn(m) is an efficient DNA cleaver[1].
We will focus our interest on modified Hoechst 33258 coupled to a metalloporphyrin[2] or on catecholate analogues of Hoechst 33258 able to chelate copper (II)[3]. In order to optimize nuclease activity of such system, investigation of interaction between these molecules and DNA is necessary.
Training content (objective, benefit and expected irnpact)
The project will involve modelling to rationnalise potential binding models of the hybrid H33258 to duplex DNA and some high field NMR studies of the complex. It may also involve fluorescence titration of the complex (including stopped-flow). I shall also be able to interact with Pr Douglas on his new DNA cleavage system, thus transferring expertise from our Toulouse laboratory.
