Genetic predisposition to skin tumor development in mice

Research objectives and content
The objectives of the project are the mapping by linkage analysis and the cloning of candidate susceptibility genes affecting inherited predisposition and resistance to murine skin tumor development in two selected murine lines. Two lines of mice (sensible and resistant to carcinogens) produced by a bidirectional selective breeding and presenting squamous cell papillomas and carcinomas will be used. Markers such as simple sequence length polymorphisms (SSLPs) and strain specific interspersed repetitive-PCR (IRS-PCR) will be mapped in the parental murine strains in order to perform the linkage analysis in the F2 progeny cross. When the chromosomal location of skin tumor
predisposition/resistance loci will be established, a positional cloning of the candidate genes will be started. Contemporary to these studies, we will begin to clone, by differential dysplay PCR and RNA substraction techniques, genes differentially expressed in the normal skin of the two strains of mice.
Training content (objective, benefit and expected impact)
As a medical geneticist, I want to orientate my career in the field of the polygenic diseases for studying their genetic basis. This training will provide me the knowledge necessary to be able to perform complex genetic and linkage analysis as well as the possibility to learn essential techniques of molecular biology for cloning new genes.
