Objective
Regulation of transcription initiation by RNA polymerase II (Pol II) requires TFIID, a multisubunit complex composed of the TATA box binding protein (TBP) and 7 to 13 TBP associated factors (TAFIIs). Coactivators which selectively mediate stimulation by different activators are associated with the TBP in distinct TFIID complexes. Recently in the host institute several new human (h) TAFIIs have been cloned and partially characterized. In the host institute, using in vitro assays several functional interactions between the identified TAFIIs and distinct transcription factors have been demonstrated, such as for example between TAFII30 and the human estrogen receptor or TAFII100 and the small subunit of TFIIF. To better understand the function of the TAFIIs they have to be studied in an in vivo system. Since in vivo studies using mammalian cells are extremly difficult and time consuming and since most of the human TAFIIs have been identified in yeast I plan to establish a genetic screening in yeast (search for supressors of ts mutants) to elucidate the role of TAFIIs in the regulation of RNA polymerase II transcription in vivo.
