Cloning and characterization of the genes regulating expression of the avirulence gene avr9 of the fungal tomato pathogen cladosporium fulvum

Objective



Recent studies with pathogenic bacteria and fungi have shown that several genes are induced when the pathogen grows in planta. The avirulence genes code for race-specific protein elicitors which are inducers of the hypersensitive response in plants with the complementary resistance genes. In order to develop durable resistance crops to pathogens by molecular resistance breeding, a good understanding of the function and regulation of avirulence genes is essential. One of these genes is the avirulence gene Avr9 of Cladosporium fulvum. In this project we will study the regulation of the expression of the Avr9 gene by performing promoter analysis and searching for cis- and trans-activating elements. Mutants will be isolated which show constitutive expression in vitro of this in planta-induced gene. Mutants with alterations in the regulatory genes will be used in complementation studies to isolate positive clones which code for the regulatory proteins.
