The "phomactins" are a new family of intriguing diterpenoid compounds which incorporates several | quaternary centers embedded in an unsual furanochroman and 12/14-membered macrocyclic containing | structure that have recently been isolated. They represent a new and exciting group of novel platelet activating factor (PAF) antagonists and are therefore attractive targets for developing a new type of antiallergic and anti-inflamatory drug.
We propose: (i) To develop synthetic methodology for elaborating the key structural features in the phomactins; (ii) To effect total syntheses of members of the phomactins, (iii) To construct a model for the biological activity of the phomactins, and make a contribution towards developing a new type of antiallergic/ anti-inflamatory drug
The key strategy to the unified synthetic approach towards several phomactins will be based on (i) elaboration of an activated cyclohexene ring precursor appropriately functionalised for (ii) an
endo-macrocyclisation in order to access the key bicyclic intermediate. Training content (objective, benefit and expected impact)
The postgraduate training allows to me learning of techniques and topics of organic chemistry which could not be achieved at home institution in Spain. Subsequently I could apply to return to home Institution or spanish industry.
