Generation of mice transgenic for killer cell hla class i receptors

Objective



Research objectives and contents
NK cells express on their surface receptors for MHC class I proteins (KIR). Engagement of these receptors inhibits NK cell cytotoxic programs. KIR can be expressed on T cell subsets and their engagement also results in inhibition of effector functions by the CD3-TCR complex. The aim of my proposal is to investigate the mechanisms of activation and/or inhibition of natural cytotoxicity with an "in vivo" model. In particular I would like to generate mice transgenic for a class of KIR, the p58 molecules and their natural mutant p50, which specifically recognize HLA-Cw3. The transgenic mice for p58/p50 molecules will be a unique model for the study of KIR role in NK cell ontogeny of the repertoire of NK cells in these mice and of the dissection of the mechanisms controlling T and NK cell activation/inhibition programs.
Training content (objective, benefit and expected impact)
My project is a part of a collaboration on NK receptors for MHC class I molecules-between the laboratory directed by Prof. L. Moretta in Italy and the one directed by Prof. E. Vivier in France. Since the Centre d'Immunologie de Marseille-Luminy has a long experience in "in vivo'' investigation using transgenic mice, I will easily conduct my project and I will be able to learn a highly specialized technique than can be very useful for my professional experience.
