The drosophila mitochondrial gamma polymerase - molecular characterization and role in the maintenance of a mtdna deletion phenotype

Objetivo



The gamma polymerase, which catalyse the replication of the mitochondrial genome (mtDNA) has been recently identified in Drosophila melanogaster and the two genes encoding the two different subunits have been cloned. The aim of the project is to study the promoter region of the catalytic subunit gene. In a first step, with classical experiments (primer extension and S1 protection analysis), We will identify putative important motifs implicated in the regulation of the gene. Once identified, they will be used to study factors which bind specifically to them. These factors will b e further characterized using the one-hybrid system in yeast.
In parallel, identification (cloning and sequencing) of the catalytic subunit gamma polymerase gene will be done in a mitochondrial mutant strain of D. subobscura in order to study a possible defect in the gene. This project will allow to extend the current knowledge of the mtDNA replication which is a main aspect of the mitochondrial biogenesis.
