Study of the role and the function of a new g protein subunit regulated pi3 kinase

Research objectives and content
My research project will be to determine the different functional domains of a new PhosphatidylInositol 3-kinase (PI3kinase). My host laboratory has isolated and cloned a new PI3kinase that is activated by heterotrimeric G-protein By subunits. This enzyme is composed
of a subunit (p120) that possesses a lipid kinase-catalytic
activity and a p101 subunit that seems to have a regulatory role. The aim of my project will be to understand the role of p101 in the activation of the PI3 kinase by G-proteins. First, I will determine the interaction domains between these two subunits and the role of this complex in the activity of the enzyme. Then, I will study the interaction with other signalling proteins. Secondly, I will study the physiologic role of this heterodimer in different cell types, such as U937 cells and Sf9 cells. Training content (objective, benefit and expected impact)
This work will involved training in Molecular Biology and lipids Biochemistry. All these techniques will be an important benefit and very useful for the development of my career as a researcher in France for INSERM or CNRS.
