Determination of genes and gene expression associated with drought resistance in hybrids between lolium and festuca species

Objective



The project aims at locating, isolating and cloning genes for drought resistance and survival transferred from Festuca arundinacea (drought resistant) into Lolium multiflorum (drought susceptible). Utilizing a novel cytogenetic procedure which is uniquely suited to these species, genes associated with drought resistance will be identified for direct incorporation in breeding programs aimed at sustainable grassland agriculture in Mediterranean areas. The exceptional ability to distinguish gene sequences within Lolium/Festuca hybrids by genomic in situ hybridization (GISH) has enabled location of Festuca genes which, in Lolium germplasm, confer extreme drought resistance Selected drought resistant lines will be used to (a) isolate and clone gene sequences from introgressed chromosome segments in Lolium and (b) identify the prime components of the molecular, genetic and biochemical mechanisms of two contrasting adaptations to drought. Characterized clones will be used as molecular markers for gene transfer in breeding and biotechnology programs
