C/EBPD is a factor believed to regulate transcription of IL-6 responsive genes and of the IL-6 gene itself and the differentiation of myeloid and Iymphoid lineages Its mRNA generates two proteins with opposite functions: the full-length trans-activator and a truncated form, LIP, thought to act as a repressor A null mutation of the gene, inactivating both C/EBPb forms causes both defective and increased transcription of different target genes. To clarify the physiological role of C/EBPB, I propose to analyse liver and macrophagic cell lines derived from C/EBPB-deficient mice and to specifically inactivate either the full-length or the truncated form of C/EBPB by gene targeting, in order to investigate the real contribution of the two forms to the functions of the factor.
Training content (objective, benefit and expected impact)
By performing gene targeting in the mouse, I will gain new experience in the study of gene expression in a complex in vivo system and learn the techniques of genetic manipulation of the mouse genome. I will also acquire more experience with derivation of stable cell lines, which will allow me a more sophisticated analysis than what is possibie in the whole organism.
