Splice-site control of a muscle-specific exon of the b-tropomyosine pre- mrna - mechanism of rna secondary structure formation and identification of trans-acting factors

Research objectives and content
Various elements have been identified that modulate splice-site selection in alternative splicing events. Among them, pre-mRNA secondary structures have been shown to activate or repress the use of specific splice sites. The b-tropomyosin pre-mRNA has provided an instructive model to study how RNA folding can control alternative splicing site usage. The muscle-specific exon has been shown to be repressed in a non-muscle cell environment by a complex mechanism that involves RNA secondary structure formation. The aim of my project will be to understand the mechanism that induces RNA secondary structure formation and to identify trans-acting factors that allow the switch of the splicing pattern towards the skeletal muscle-specific form during myogenic differentiation.
Training content (objective, benefit and expected impact)
After having studied RNA structure-function relationship in RNA phages, this project will give me the opportunity to study RNA structure-function in more complex organisms.
