Objective
Research objectives and content
The Patched molecule is a novel membrane receptor for the morphogen protein hedgehog. In man Patched acts as a tumor supressor. Probably Patched controls the activity of the morphogen Smoothened. The effect of Patched and Smoothened on keratinocyte development shall be studied. Repression respectively hypertrophy of the follicular epidermal cells are expected however the effect of these alterations on follicle architecture are as well a point of interest since they reflect the pathways of cellular communication between the different types of cells involved. A second part of the project will be the signalling cascade downstream of Smoothened. Despite of considerable knowledge about the effect of patched on the cellular level the the direct effect of Smoothened is not yet revealed. Using the two-hybrid system the intracellular ligand(s) of Smoothened should be identified.
Training content (objective, benefit and expected impact)
The most actual techniques in modern biology will be applied (transgenic animals, functional gene library screening). The studies will reveal a so far not investigated pathway of intracellular signalling and also provide inlportant information about embryonic skin development and about the most frequent skin tumor.
