Fish gill band 3-like anion exchanger - ae2

Objective



The objective of the proposed study is to determine the distribution of band 3-like protein (AE2) in the gills of aquatic vertebrates. The distribution of AE2 will be determined using immunocytochemistry at the light and electron microscopic levels. A suitable antibody probe will be developed against fish erythroid band 3. I will purify the cytoplasmic fragment of band 3 protein from teleost (Oncorhynchus mykiss) red blood cells under non denaturing conditions using differential centrifugation and ion exchange and size exclusion chromatography. Antibodies will be raised against this purified peptide and once the specificity has been validated used to determine the cellular distribution of AE2 in the fish gill under different environmental conditions. In this way the function of different cell types (acid, base excreting cells) in this acid-base reguatory epithelium can be determined.
