Investigation of growth factor production essential for the development of human hematopoietic stem cells - 'hsc' - from muscle precursor cells

Objetivo



Research objecuves and content Among manv limiting elements of current protocols of somatic gene therapy, the delivery of growth factors that are essential for the proliferation of hematopoietic stem cells (HSC), is an important aspect. The aim of this training project is to establish an improved animal model for the development of human HSC. I propose to deliver via muscle cells, growth factors that are important for HSC development. I plan to construct genes for the production of growth factors under tetracycline inducible regulatory promoters. The constructs will be transformed into conditionally immortalized muscle precursor cell lines developed in the host lab. These cells nvill be reimplantated into the limbs of mice to form normal muscle cells with the capacity to deliver growth factors. The growth factor production of these muscle cells will be tested in vitro and in vivo Training content (objective, benefit and expected impact) My hostess Dr. Jenny Morgan. who works in the muscle cell biology group in London. headed by Dr. Terry Partridoe. established the lines and the culture conditions for the myogenic precursor cell lines. the experimental transformation conditions and the graft methods. Durino my training period. I will learn these skills and after vards return to Giessen in order to establish these methods in the home institute. Links with industry / industrial relevance (22) The results obtained durina this training period will be important for establishing an animal model that supports the development of human HSC with the production of essential growth factors. The engineered mice can be profitable to the industry as well as academic community for further studies of HSC with respect to gene therapy of human lymphapoietic disorders.
