Synthesis of antimitotic agents related to rhazinilam

Research objectives and content
Synthesis and structure-activity relashionship of rhazinilam and analogs. The project will focus on the biaryl structural element of the natural product. A strategy involvino heterocouplinn of two highly functionalised aromatics will be used. This strategy is meant to be used in a combinatorial approach on a so- lid support. The biological evaluation will be done by the researchers of Rhtne- Poulenc Porer in Paris. With these data in hands we hope to be able not only to design better analogs but to understand the mode of action of rhazinilam which is still under debate. Also this project will require to develop new practical routes towards heterobiaryls and will help the project of total synthesis of rhazinilam itself which is presently studied in Ghosez's laboratory.
Training content (objective, benefit and expected impact)
The objective is to train a Ph.D. student in the field of organic synthesis. This project is particularly interesting inasmuch as it involves synthetic methodolo- gy, combinatorial synthesis and structure-activity relationships.
Links with industry / industrial relevance
The link will be strong because the biology will be done in industry. Obviously this fundamental research project is a great interest for the pharmaceutical industry because rhazinilam acts as an antimitotic agent like taxol and taxotere which are powerful anticancer agents.
