Mitochondria is the major cellular source of reactive oxygen species (ROIs) such a hydrogen peroxide, superoxide anion and hydroxy radical. Nevertheless, mitochondria lacks catalase and superoxide dismutase, two main enzymatic antioxidant defenses in the eukaryotic cell. A novel thioredoxin has been identified in mitochondria (Trx2) which could be involved in scavenging ROIs. The aim of the proposed project is the fully characterization of the mitochondrial thioredoxin system which comprises the identification of the function(s) of Trx2, regulation of the gene expression and the isolation of the mitochondrial thioredoxin reductase,. Training content (objective, benefit and expected impact)
The proposed project will provide the applicant a complete training in most of the molecular and cellular biology techniques. Additionally, many important diseases like Alzheimer, Parkinson, rheumatoid arthritis, etc., have been directly related to mitochondrial disfunction and an altered production/inactivation of ROIs. The characterization of the novel mitochondrial thioredoxin system as a main antioxidant defense and its possible role in the treatment of these diseases could be thus of major interest.
