Characterization and analysis of a family of uncoupling proteins. Importance for metabolic diseases

Research objectives and content
- Study of the human Uncotll)lillg Protein-2 gene to determine the organisation of the gene, its possible polymorphism and its relationship with diseases related to the energy metabolism as diabetes, obesity, etc. - Analysis of the biological roles of the Uncoupling Protein-2 on the Resting Metabolic Rate, its relation to the effects in energy metabolism action of several hormones as thyroid hormones. - Study of the possible role of the Uncoupling Protein-2, which is also expressed in macrophages, spleen, thymus and lymphocytes, in the mechanism of inflamation immunity and the production of fever. -Search of other components of the family of Uncoupling Proteins, based on the cloning of a murine uncoupling protein mostly expressed in skeletal muscles Study of its capacity to uncouple respiration.
Training content (objective, benefit and expected impact)
This project will provide the applicant with background expertise to work with molecular biology techniques that constitute the basic step in the development of the work Several techniques involving gene cloning, sequencing, Southern, Northern and immmuno-blotting, plasmid construction and transient transfection of adypocite-derivated cell lines experiments, will be used as global approach to the study of gene organisation and function. The majority of these techlliqlles are in routine operation within the host institution (Centre de Recherche sur l'Endocrinologie Moleculaire et le Developpement), therefore the applicant should obtain definitive results within the postdoctoral stay.

The relation of Uncoupling Protein-2 with the resting metabolic rate as well as the diet induced thermogenesis strongly suggests that it may be target for new obesity-therapy compounds Major pharmaceutical companies should be interested in research on UCP2 and other UCP homologues.
