Identification of natural substrates and subunit specificity of the thermosome, a group ii chaperonin from the archaeon thermoplasma acidophilum 6

Research objectives and content
Chaperonins are high molecular mass proteins ubiquituos in a variety of cellular compartments and assist the correct folding and assembly of polypeptides in an ATP-dependent manner. In archaea, the chaperonin family is represented by thermosomes. This project is aimed to investigate some important aspects concerning the thermosome from Thermoplasma acidophilum identification of its natural substrates, establishment of folding assays and study of subunit specificity. Different biochemical and molecular biology techniques will be used and the results complemented with some other approaches which are being carried out in the research group. Training content (objective, benefit and expected impact) The objective of the training is to learn different approaches to protein study and it will complement my previous training on protein structure spectroscopy. The expected results will be of great importance to the advance in the functional and structural studies of chaperonins.
