Objective
This project will offer postdoctoral scientists the opportunity to acquire knowledge and experimental expertise from synthetic organic chemistry to occurring at the active sites of coenzyme B12-dependent enzymes. This will be achieved by an interdisciplinary approach using the complimentary skills molecular biology. Such interdisciplinary training is of increasing importance to the European pharmaceutical industry, enabling it to maintain of seven leading European laboratories. The combined B12 group will be the its competitive edge.

* Structure determination of B12-dependent enzymes and protein fragments * Preparation of modified enzymes by site directed mutagenesis; evaluation of their catalytic efficiency
* Synthesis of coenzyme analogues, preparation of enzyme-coenzyme analogue complexes and evaluation of their catalytic efficiency
* Synthesis of substrate analogues and evaluation of their inhibitory properties
* Characterisation by spectroscopic techniques (NMR, EPR) of enzymes and enzyme-coenzyme complexes, in the presence and absence of substrates and inhibitors
* Development of functional models for B12-dependent enzymic reactions. The project builds on a current successful collaboration of four of the laboratories, which has provided new insights into the B12 mechanistic problem.
