Objectif
The description of the molecular basis of the activity of biological molecules is deeply linked to the knowledge of their three dimensional structure at the atomic level. This is why a lot of effort has been dedicated to the development of techniques, which allow the determination of the structure of biomacromolecules. In the last few decades, NMR spectroscopy has emerged as one of the most powerful tool for the study of the structure and the dynamics of biological macromolecules. It determines about 25% of all new three-dimensional structures and is the only technique capable of providing high resolution structural information on these molecules in aqueous solution.
The two proposed meetings of this series "NMR in Molecular Biology" will be focussed on Structural Genomics and NMR. Structural Genomics aimed at using high-throughput structure determination and computational analysis to provide three-dimensional models of every tractable proteins. Several pilot projects in Structural genomics are developing over the world and of course in Europe and it is important to emphasise now the specific role of NMR and its complementarity with X-ray studies to increase efficiency and rapidity in structure determination but also to establish stronger links with function. The expected technical improvement from Structural Genomics will and is already transforming Structural Biology, as solving protein structures will become easier. Thus Structural Biology will have to turn to more demanding projects, for example solving the structure of bigger objects or membrane proteins and to be more deeply involved into Biology.

In the first meeting it is proposed to present the main projects in Structural Genomics under development, the number of structures solved as well as a survey on how expression, purification and labelling of these molecules have been handled and improved efficiently. Then, the methodological aspects raised with rapid structure determination will be tackled. Finally, the problem of badly or not completely folded molecules will be addressed in the protein folding session. The second meeting will follow up on the development of NMR in structural biology, and particularly with its role in structural genomics in terms of function and molecular interactions. The emphasis may be even more shifted towards new aspects and new methods to study dynamics and interactions between biomolecules to fully understand their functions. Of particular interest will be the membrane protein structure determination both by solid state NMR and high-resolution liquid NMR as these usually form the majority of drug targets. The topics and the speakers selected for the two meeting are complementary allowing a full coverage of the field.
