Identifying novel regulators of the eukaryotic cell cycle by functional genomics

Objective

The aim of the project is to take an approach based on functional genomics to identify novel regulators of the eukaryotic cell cycle. The molecular mechanisms that regulate cell cycle progression are conserved throughout the evolution of eukaryotic organisms, from yeast to mammals. Therefore, we propose to identify novel cell-cycle regulators, by analyzing the function of a large set of uncharacterized genes in Saccharomyces cerevisiae. Budding yeast possesses 6000 genes in the genome and around one third has no function established. There have been described all the essential budding yeast genes under laboratory conditions. We are planning to analyze all the yeast genes that are essential and are uncharacterized experimentally, by constructing temperature sensitive degron mutants. Around 200 yeast genes meet these criteria. Many of these genes have homologues in higher eukaryotes, so that the results in budding yeast are likely to be of importance to provide a model for the function of homologous genes in higher eukaryotes.
