Bacterial chromosome partitioning: the role of streptomyces para and parb proteins

Objective

In contrast to previous views, recent studies have shown that bacterial chromosome partition is active process. All studies done so far have focused on unicellular bacteria with circular chromosome. However, they revealed that the functional significance of partition proteins differs for different bacterial species. This proposal focuses on partitioning of the Streptomyces linear chromosome during the complex life cycle of this mycelial organism. Recently identified Streptomyces chromosome partition genes, parA and parB and their products, ParA and ParB, will be further characterized, leading to an analysis of the role of these proteins during partition complex formation and regulation of their expression during Streptomyces growth and development. Putative mechanism of coupling chromosome replication to partition involving the replication initiator protein (DnaA) will be explored. The following aims of the research project are planned:
(i) analysis of interactions of ParA/ParB,
(ii) the sub cellular localization of ParA and ParB as well as DnaA, particularly in mycelia compartment undergoing multiple cell division, and
(iii) study of developmental regulation of transcription from parAB promoter. The proposed study should provide insight not only into Streptomyces linear chromosome partitioning but also into its correlation with DNA replication and colony sporulation.
