Objective
The main objectives are:
1) To study the effects of temperature (T) on carbon (C) metabolism and translocation in arbuscularmycorrhizas (AM), to determine if low temperatures limit the development of the extrardical mycelium to a larger extent than the intraradical mycelium due to a reduced mobility of lipid bodies, a decrease in C transfer from the plant, in the synthesis of triacylglycerides, or in the formation of lipid bodies; and
2) To explore C limitation in the nutrition of AM fungi to determine if
i) plant C supply precludes at any time AM fungi development or function and
ii) AM fungi's response depends on their C and N status and/or developmental stage.
The effects of T and CO_2 on C nutrition and metabolism in different isolates of AM fungi will be studied in 3 experimental systems simultaneously scaling up in complexity and reality: monoxenic root organ cultures, monoxenic whole plant cultures and plant-soil microcosms, where C will be followed by means of microscopy and microscopy data analysis, stable isotopic C labelling followed by nuclear magnetic resonance (NMR) and biochemical analysis of lipids, and radioactive C labelling and scintillation counting. This information is crucial for accurate prediction of C movement into the soil and realistic C budgets.
