Objective
The terms "induced accessibility" (IA ) and "induced inaccessibility" (ll) have been adopted to describe situations where early attacks increase susceptibility or resistance, respectively , to later attacks. II is extremely effective, race non-specific and persistent. However the molecular basis of IIis unknown. If we can understand the basis of induced changes, we may be able to modify the plants intrinsic ability to resist disease by enhancing constitutive expression of factors that are normally expressed only after induction of inaccessibility, The current proposal aims to identity the basis ofII in relation to powdery mildew, one of the most damaging diseases of temperate cereals. The primary objective will be to understand the biochemical basis of II to B. graminis by using chemical inhibitors to disrupt host metabolic pathways known to influence resistance (AOA,OH-PAS, DDG, Mannose, Cicloheximide and Cytochalasin. Additional experiments will examine whether oat crown rust attack (on mespophlly cells) induces changes in epidermal cell response to powdery mildew attack. This will explore the possibility of cross-talk inducing changes between different plant leaf tissues. The applicant will benefit directly from acquiring a detailed understanding of the biology of host resistance and Jeaming biochemical and histological analytical procedures to elucidate complex interactions using light and electron microscopy. Whatever the outcome of individual experiments using inhibitory chemicals, the results will definitely provide valuable information on the basis of induced inaccessibility.
