Elucidation and characterisation of epithelial cell phenotypes

Objective

A key factor in the pathogenesis of several clinically important diseases is epithelial cell dysfunction. This proposal will investigate novel areas of epithelial cell biology with the aim of enhancing our understanding of the role of epithelial cells in the development and progression of chronic dermal wounds, COPD/asthma and endometriosis. Project 1 will investigate the role of retinoid receptor agonists on epithelial cell phenotype during dermal wound healing. The project will characterize retinoid receptor expression in primary human keratinocytes before and after mechanical injury. Subsequently, the study will investigate retinoid mediated gene expression in both in-vitro and in-vivo wound models. Project 2 will study the role of estrogen on endometrial cell function at a molecular level. This project will identify biomarkers of estrogen function in endometrium in-order to expand understanding of how estrogen mediated gene regulation impacts on endometrial estrogen response. Project 3 will investigate the responses of lung epithelial cells following allergen or bacterial challenge in-vitro. The project will identify changes in gene expression after exposure to house dust mite and H. influenzae, and investigate links with the release of key cytokines. Projects will employ state of the art micro-array technology (which is not available in most academic institutions) in-order to develop understanding of epithelial cell gene expression in a number of biological paradigms.
