Nmr tools for the study of transient intermediates in biomolecular processes

Objective

Objectives: The proposal aims at the development of dedicated NMR probes and the accompanying NMR tools for the structure determination of rapidity formed, short-lived intermediates that are important in biological processes. These transient intermediates can be induced in a biomolecular system by photochemical means or by a rapid change of solvent conditions. Therefore, both a rapid mixing and a laser coupled probe will be made in which all modern multidimensional NMR experiments including the use of pulsed field gradients are possible, with as little compromise on the NMR performance as possible. These NMR probes and the set of developed experimental methods will be validated for photobiology, in protein and RNA folding studies, and in CIDNP experiments.
