Objective
The main objectives are as follows :
* the further development of DNA markers for coconut with specific emphasis on STM, ISTR, AFLP, RAPD and RFLP markers;
* to increase the marker density on the first coconut map (1997) established in the frame of an EC STD3 project (1/1994-4/1997);
* to analyze available coconut germplasm for resistance or tolerance to biotic stress;
* to identify QTL markers for other important traits (for example dwarfism or early- bearing);
* to examine the usefulness of coconut markers in other economically important Palmae like the oil palm (2n=32; synteny of genomes for coconut / oil palm);
* to develop appropriate software for assistance in QTL analyses; and
* to make public the project's progress on its coconut home page (http : //www.mpiz-koeln.mpg.de/~rohde/coconut.html) as well as disseminate results and expertise in annual workshops / laboratory courses in the coconut-growing regions (South East Asia / Pacific; Africa; Latin America / Carribean).
Expected Outcome
The work carried out in this project should assist breeding in coconut by providing the basis for biodiversity analysis of coconut germplasm through DNA marker technologies. Further assistance is provided by establishing a coconut linkage map incorporating a variety of DNA marker types and integrating 3 different mapping populations established from local as well as coconut germplasm used widely in breeding programmes. One important aspect is the dissemination of results and technical expertise in combined annual workshop / laboratory courses.
The key activities envisaged are :
* The identification of codominant RFLP and STM markers by cloning, sequencing and polymorphism analysis and their integration together with RAPD markers into the existing ISTR / AFLP coconut linkage map.
* While RAPD, ISTR and STM markers will be used by DC partners in local and introduced coconut germplasm analysis as well as in mapping efforts with their respective mapping populations, AFLP and RFLP will be developed by MPIZ and CIMA specifically as markers for the coconut linkage map. Both European laboratories will adapt AFLP and ISTR to the use of fluorescence-labeled PCR primers for automatic large-scale analyses on sequencing machines.
* The mapping populations will serve as unselected populations in attempts to identify DNA markers that would segregate with quantitative traits like early flowering. Further activities will be devoted to the identification of markers for the Makapuno endosperm phenotype, dwarfism and possibly tolerance to phytoplasm.
