Objective
Detection of biomolecules using fluorescent probes is a cornerstone of up-to-date cancer diagnostics and research. The necessity to simultaneously measure a greater number of biological indicators imposes new demands on the probes for multicolour experiments, which should emit at spectrally resolvable energies and have narrow, symmetric emission spectra, and the entire group of probes should be excitable at a single wavelength.
This project consists of developing a new approach to fluorescent analysis of biomolecules based on their bioconjugation with semiconductor NCs, which are 100-fold more photostable than common fluorescent dyes and emit a narrow (20-30 nm) fluorescence spectrum with a quantum yield as high as 70%. Varying the material used for NCs and size of the NCs allows one to obtain a spectral range of 400 nm to 2? m in the peak emission and large extinction coefficients in the visible and ultraviolet ranges. NCs of various sizes may therefore be excited with light of a single wavelength, resulting in many emission colours that may be detected simultaneously. The project will be carried out by six research teams with an expertise in various areas, such as inorganic, physical, and bioorganic chemistry; physics, biophysics, immunochemistry, genotyping and cancer research.
This three-year program has five objectives:
(i) to synthesize homogeneous in size (5% variation) ZnS-capped CdSe nanocrystals with diameters ranging from 3 to 7 nm, emitting fluorescence in the range from 500 to 650 nm with a quantum yield >50%;
(ii) to modify the surface of NCs with organic surfactants and to analyse the effects of surface activation on the photoluminescence properties, donor/acceptor characteristics, and photo- and chemical stability of NCs in buffer solutions with different ionic strengths and temperatures;
(iii) to perform bioconjugation of the nanocrystals with antibodies against DNA topoisomerases I and II, exciting drug targets (in clinical oncology), and with DNA and oligonucleotides to be used in molecular beacons, hybridisation probes that ensure rapid, highly sensitive, and inexpensive genotyping with a base-pair accuracy;
(iv) to adapt and apply the bioconjugates to quantitative detection and/or imaging of DNA topoisomerases by confocal microspectroscopy, cytofluorimetry, or the standard immunoassays; and
(v) to apply the nanocrystal-based molecular beacons to hybridisation of DNA with its complementary targets in homogeneous solutions.
The goal of the project is development of new ultra-sensitive non-isotopic assays based on the use of fluorescent nanocrystals. The nanocrystals may be used for labelling of the tumor precursors in clinical diagnostics, visualization of antibody-antigen binding, and real-time genotyping, analysis of single nucleotide polymorphism, and in multiplex real-time polymerise chain reaction.
