Objective
Methods for the genetic manipulation of the unicellular, N2-fixing cyanobacteria Gloeothece and Synechocystis will be developed. The aim of the project is to produce modified strains that can be used in physiological and biochemical studies of the interactions, at the molecular level, between N2 fixation and O2.
The following work will be undertaken: isolation of appropriate mutants of these cyanobacteria; development of a transformation system that can be used to introduce novel genes into these organisms; and construction of genomic DNA libraries for Gloeothece ATCC 27152 and Synechocystis BO 9201.
