Structural and functional properties of molten globule state of proteins

Objective



The physical state of protein molecules known as the molten globule state has been discovered and extensively characterised at the Institute of Protein Research, Pushchino, Russia. It has been proposed, or proved on an experimental basis, that this state plays a physiological role in several processes.

This collaborative project will consist in the study of: the influence of conditions that mimic the environment near membranes on the physical state of retinol-binding proteins; the role of structural transitions in the process of targeted delivery of vitamin A derivatives to physiological acceptors; and the structural features of the molten globule state of all proteins of distinct topologies, ranging from all helical to all sheet proteins, including retinol-binding protein, cellular retinol-binding protein, lysozyme, a-lactamase, as well as recombinant chimeric proteins.

The methodologies that will be used include high resolution NMR, UV circular dichroism, fluorescence, IR spectroscopy, microcalorimetry, hydrodynamic techniques and X-ray crystallography.
