Crystallographic analysis of a presynaptic glutamate receptor and molecular cloning of trans-acting factors and cis-acting elements by genetic selection in yeast

Objective



The first part of this project is directed to the structural study of a new family of presynaptic proteins cloned by CNRS. This new family of presynaptic proteins forms a glutamate neuroreceptor when expressed in vitro. Methods will include a construction of neuroreceptor when expressed in vitro; a construction of recombinant plasmids based on yeast expression vectors and purification of the proteins. Further crystallographic analysis of the protein structure will be performed at the Medical Research Council Centre in the United Kingdom.

The second part of the project is in line with previous studies performed at CNRS,France, and concerns the study of the promoter region of the gene coding for a neurotransmitter biosynthetic enzyme tyrosine hydroxylase (TH). The project will be directed to the cloning of neuronal transcription factors, which recognise specific sites of the TH promoter, by using a collection of yeast mutants and expression vectors. Secondly, the techniques developed at CNRS and at St. Petersburg University, will allow identification and isolation of different members of the multimetric protein complexes involved in TH promoter activity.
