Objective
The aim of this project is the development of new allosteric systems which by suitable conformational coupling between heterotopic binding sites I and II should show exceptional degrees of co-operativity. The following will be used as conformational coupling units: cyclohexanes equipped with crown ethers as binding sites for alkali, ammonium ions etc., and substituents R suitable for organic substrates including anions etc.; bicyclononanes with 2 nitrogen atoms as soft metal binding sites, and either crown ether or larger rigid substituents, the latter aimed at stacking interactions with aromatic substrates; and calixarenes with soft binding sites at the upper rim and hard binding units at the lower rim.
Evaluation of the complexation properties will be carried out mostly in Chernogolovka, partially also in Saarbrücken where more physico-chemical techniques will be applied. The project requires the application of sophisticated techniques such as NMR NOE and shift evaluations, computer-aided molecular modelling, evaluation of multistep-equilibria.
The new systems with well-defined conformational mobility should open the way to systems suitable for many future applications, such as controlled ion complexation and transport, release of toxins, antitoxins or catalysts. Analytical applications could extend to the simultaneous determination of combinations of hard and soft cations. The systems therefore are expected to be of considerable scientific and medicinal as well as industrial significance.
