Development of early markers for detection of environmental stress in aquatic organisms

Objective



Objectives and Research Activities:
This multidisciplinary project is concentrated on the development of novel mollar biological techniques for monitoring of the quality of aquatic environments. The effect of environmental stress on freshwater sponges and algae will be studied using the following biomarkers of exposure: (i) heat shock protein HSP70 and DnaJ protein homolog, and (ii) polyphosphates (polyP). This will enable us to establish an early warning system to detect environmental perturbation in aquatic ecosystems before irreversible damage occurs.
The measurable, specific objectives of the proposed research are: To determine the expression of stress proteins (heat shock protein HSP70 and DnaJ protein homolog) in sponges. Specific tasks are: (a) To assess and to characterize the effects of different doses of known pollutants on sponges. (b) To establish sponges as a model systems for testing environmental stress in the field. To investigate the role of polyP in stress response of algae, in complex formation with metal ions present in polluted aquatic environments and in the development of algae blooms. Specific tasks are: (a) To investigate synthesis and degradation of inorganic polyP during stress response of algae to changes in
(i) pH,
(ii) osmolarity and
(iii) phosphate concentration. (b) To investigate function of inorganic polyP in complexing heavy metal ions (Cd, Cu, Zn, etc.) present in polluted freshwater. Study sites:
(a) Lake Baikal (Russia) and
(b) Lake Kinneret (Israel). These lakes are utilized simultaneously as sources of domestic, industrial and agricultural supply of water, fishing grounds, drainage basins of sewage as well as recreation sites. Consequently, they are constantly under a man-made threat of disturbance of their ecological equilibrium. Expected results:
Availability of cDNAs probes for HSP70 and DnaJhom from the selected freshwater sponge species (Lake Baikal); availability of monoclonal antibodies for HSP70 and DnaJhom from the selected freshwater sponge species (Lake Baikal); elucidation of the level of these proteins in sponge organisms living in different polluted areas; development of an ELISA system for quantification of HSP70; development of an ELISA system for quantification of DnaJhom; characterization of polyP kinase and exopolyphosphatase(s) from algae; elucidation of the effect of stress on polyP metabolism in algae; elucidation of the role of polyP in complexing heavy metal ions.
