Mechanisms of cellular adaptation to ionising radiation

Objective



The research to be carried out aims to investigate the mechanisms by which cells can cope with such situations by adapting their repair capability and to define the factors which play a role in such adaptive responses. Earlier studies in the laboratory of one participant have shown that an adaptive response resulting in a reduction of genetic damage can be elicited also in unconditioned cells by means of a factor present in, or released by, cells subject to a conditioning irradiation. The experiments planned in the proposal presented will define more clearly the release of the transmissible factor with respect to dependence on time, doses of conditioning and challenge, the nature of the transmission - by substances released from pre-irradiated cells or direct contact between cells-, the nature of the factor - size and type molecule (macromolecular, protein) -, the role of the factor in the reaction to other agents (mutagens) and the possible significance in vivo - in people treated with radiation or living in areas of high radiation exposure This observation of a transmissible factor involved in eliciting an adaptive response has not yet been described by other laboratories and is of great importance not only for our understanding of the mechanisms of the adaptive response but, once this factor is isolated, might also be useful for preventive treatments to render persons to be exposed more resistent against damage and/or to recognise persons which had been exposed to conditioning circumstances in contaminated areas. Such basic research performed in cooperation with institutes in the European Union will develop expertise in two laboratories in Kazakhstan which now are already active in the field. This will help them to face the specific problems present in Kazakhstan and to study genetic damage and its reDairabilitv in DeoDIe livina in contaminated reGions..
